The project aims at developing 3D polymer culture systems to control expansion and differentiation of human hematopoietic stem and progenitor cells (HSPC) ex vivo. HSPC-supportive decellularized ECM preparations derived from primary mesenchymal stromal cells (MSC) will be investigated with RNA-interference to identify the molecular determinants of cell adhesion, migration and expansion. MSC culture conditions and matrix processing schemes will be varied to adjust the characteristics of the obtained matrices. In parallel, we will customize a set of in situ assembling poly(ethylene glycol)-heparin hydrogels by adjusting elasticity, enzymatic degradability and presentation of ECM-derived peptides and growth factors for the defined 3D stimulation of embedded HSPC. Cells expanded under the most effective conditions will be tested for their early homing and long-term engraftment potential using a xenograft model.
