Abstract:    Epigenetic regulators such as histone methyltransferases and demethylases have been identified as critical regulators for development. We have studied UTX - a histone 3 lysine 27 specific demethylase – and demonstrated its critical role during embryogenesis as well as for normal hematopoiesis and leukemic stem cells. Moreover, our results suggest a histone-demethylase independent function of the epigenetic regulator UTX and its male counterpart UTY. To gain insights into histone-demethylase dependent and independent functions, we will study the intracellular localization of UTX and UTY, analyse interaction partners and compare their influence on DNA replication as well as repair in primary specimens from single and double knockout mice of UTX and UTY.
