Abstract:    This Gerok project aims to optimize the in-vitro differentiation of chromaffin stem cells before these can be used for neuro-transplantation in a rat model of Parkinson disease. Potential chromaffin stem will be isolated from the adrenal medulla and subjected to treatment wit

h various agents for different times to induce dopamine production and differentiation into neural cells. The suitability of the cell for transplantation will be judged by systematic gene expression profiling, expression of differentiation markers, morphological features and eventually the relieve of extrapyramidal symptoms after transplantation into rats suffering from Parkinson-like symptoms. Although proof of principle for the differentiation of chromaffin stem cells into dopamine producing cells has already been obtained, a systematic optimization of the cell differentiation protocol is likely to benefit the currently funded and ongoing work within the DFG network project. It would also provide more insight into the differentiation potential of chromaffin stems cells. In addition, the project may better define the transformation potential of the transplanted cells which might be a safety concern if similar approaches will be tried for the treatment of patients suffering from Parkinson disease.
